Unless you are a glutton for punishment, I'd suggest you don't remove the
toilet unless you are certain there is a problem.  I've been there, done
that and have the greatest respect for the job.  It's no fun and will wipe
out an entire day. 

Instead, why don't you just reach down there and feel of the flange? It
would be profoundly easier way to get the data you want - especially if you
don't smell any odor today and if you don't see any seepage today.

With a flashlight, look down the toilet's throat and locate edges of the
flange (below the tube). Then, coat your "small" hand with KY jelly (water
soluble), reach down and run your fingers around the flange.  It's easy to
determine if the flange is loose or if it is firmly in place. Be ready to
put more KY jelly on your wrist before pulling your hand back out. The
tube's edge can be sharp on the way up.
